What Is a Certificate of Analysis?
A Certificate of Analysis (COA) is an official document that reports the results of analytical testing performed on a specific batch of a research compound. It serves as the definitive proof of identity, purity, and composition — the laboratory equivalent of a birth certificate for your peptide.
Key Components of a COA
HPLC Purity Analysis
High-Performance Liquid Chromatography (HPLC) is the gold standard for peptide purity determination. The HPLC trace on your COA shows:
- Retention time: The time it takes for the peptide to elute from the column, characteristic of the specific compound
- Main peak area percentage: The primary metric — your purity value (e.g., 98.5%)
- Impurity peaks: Smaller peaks representing truncated sequences, deletion products, or oxidation byproducts
A purity of ≥98% indicates that the main peptide species constitutes at least 98% of the total integrated peak area.
Mass Spectrometry (MS)
Mass spectrometry confirms the molecular weight of the peptide. The MS spectrum displays the observed mass, theoretical mass based on the amino acid sequence, and mass accuracy in parts per million (ppm). A mass within 1-2 Da of the theoretical value confirms correct sequence synthesis.
Physical Appearance and Storage
The COA documents the peptide's physical form — typically a white to off-white lyophilized powder. Reputable suppliers include reconstitution guidance and storage recommendations based on the peptide's sequence properties.
How to Verify a COA

- Confirm identity: Mass spec data should match the expected molecular weight
