The Science of Lyophilization

Lyophilization, or freeze-drying, removes water from a frozen peptide solution under vacuum through sublimation. This technique preserves peptide structure and activity by eliminating the aqueous environment required for hydrolysis, oxidation, and microbial growth.

A properly lyophilized peptide appears as a white to off-white powder at the bottom of the vial. Any discoloration — yellowing, browning, or darkening — indicates degradation.

Short-Term Storage (Days to Weeks)

  • Lyophilized form: Store at -20°C in a desiccated environment. Keep vials sealed with parafilm to prevent moisture ingress. Allow vials to reach room temperature before opening to prevent condensation.
  • Reconstituted form: Store at 4°C for short-term use (1-2 weeks). Divide into single-use aliquots at the time of reconstitution to minimize freeze-thaw cycles.

Long-Term Storage (Months to Years)

  • Lyophilized peptides: Store at -20°C to -80°C. Lower temperatures extend shelf life. Avoid frost-free freezers, which cycle through warming phases.
  • Reconstituted peptides: Aliquot and store at -80°C. Flash-freezing in liquid nitrogen before transferring minimizes ice crystal formation.
  • Moisture exclusion: Use desiccant packs. Peptides are hygroscopic and absorb atmospheric moisture.

Reconstitution Guidelines

Solvent Selection

  • Hydrophilic peptides: Sterile water, PBS, or saline
  • Hydrophobic peptides: May require DMSO or acetonitrile initially, then dilution into aqueous buffer
  • Cysteine-containing peptides: Use degassed buffers to prevent disulfide bond formation

Step-by-Step Protocol

  1. Warm the lyophilized vial to room temperature (15-20 minutes)
  2. Calculate the required solvent volume based on desired concentration
  3. Add solvent slowly down the vial wall to avoid foaming
  4. Gently swirl — do not vortex or shake vigorously
  5. Allow to sit for 5-10 minutes for complete dissolution
  6. Inspect visually — solution should be clear with no particulates
  7. Aliquot immediately into single-use portions
  8. Label each aliquot with peptide name, concentration, date, and solvent

Common Storage Mistakes

  • Leaving vials at room temperature — peptides degrade exponentially faster
  • Repeated freeze-thaw cycles — each cycle causes cumulative damage
  • Using frost-free freezers — the defrost cycle raises temperatures periodically
  • Moisture contamination — always cap vials tightly
  • Vortexing reconstituted peptides — mechanical stress can denature sensitive sequences
