The IGF System: An Overview

The insulin-like growth factor (IGF) system comprises two ligands (IGF-1 and IGF-2), two receptors (IGF-1R and IGF-2R), six IGF-binding proteins (IGFBP-1 through -6), and several IGFBP proteases. This intricate regulatory network controls cell growth, differentiation, survival, and metabolism across virtually every tissue type.

IGF-1R is a tyrosine kinase receptor structurally similar to the insulin receptor. Ligand binding triggers receptor autophosphorylation, recruitment of IRS-1/IRS-2 adaptor proteins, activation of PI3K/Akt pathway (cell survival and protein synthesis), and activation of Ras/MAPK pathway (cell proliferation).

What Makes IGF-1 LR3 Different?

IGF-1 LR3 (Long Arg3-IGF-1) is a synthetic analog of human IGF-1 with two modifications:

  1. Arginine substitution at position 3: Replacement of glutamate with arginine reduces binding to IGFBPs by approximately 100-fold
  2. N-terminal extension: A 13-amino-acid extension further reduces IGFBP affinity

The net effect is a peptide that is functionally more potent than native IGF-1 because it remains unbound and bioavailable rather than sequestered by IGFBPs. In research settings, this means lower effective concentrations, more predictable dose-response relationships, reduced interference from endogenous IGFBP expression levels, and longer half-life in culture.

Receptor Binding Profile

IGF-1 LR3 maintains high affinity for IGF-1R while showing minimal cross-reactivity with the insulin receptor — important for research applications where insulin receptor activation would introduce confounding variables.

  • Cell proliferation assays: C2C12 and L6 myoblast lines, primary fibroblasts, neural progenitor cells
  • Differentiation studies: Myogenic, osteogenic, and chondrogenic differentiation
  • Survival/anti-apoptosis assays: Protection against serum-withdrawal-induced apoptosis

Storage and Handling

Store lyophilized at -20°C. Reconstitute in 10mM HCl or 100mM acetic acid initially (IGF-1 LR3 is poorly soluble at neutral pH), then dilute to working concentration in assay buffer. Do not store reconstituted solutions — aliquot and freeze at -80°C.
